I also have this issue with my Roku Voice Pro second edition remote. I have tried to unpair and re-pair the remote.

Hey I posted a while ago and someone asked me for info and since then nothing. Pretend to help them disappear. Should have never bought this stupid version 2. Buy the old voice remote this new ones junk doesn't work waste of money if u want voice function

If the mic button on the Roku Voice Pro (2nd Edition) isn’t working, try rebooting the device, checking remote settings, or re-pairing the remote. Also, ensure firmware updates are installed. For Deezer Mod APK users, keep app updates current to avoid functionality issues and enjoy uninterrupted music streaming.
I was sent a new remote to "solve the issue". The new remote has the exact same problem. The microphone button does nothing.
