I have a UHD monitor with HDMI ports, but no HDMI ARC or optical input. Will the Roku streaming bar play the audio directly since the audio comes from the bar in the first place?

Why does the audio have to come back to the streaming bar from the monitor?

Please be aware that in order for your Roku streaming bar to work with audio, it should always be connected to an HDMI ARC input.
