If a private channel is already published but there are new changes made, is it as simple as uploading the new package from the developer site? I have a saved copy of the current package of the private channel.

Made changes to the graphics, etc and need to upload the changes to test.

Yes, for private channels just upload your changes, test with the Access code and then hit "Publish" when you are sure it is ready, use the Vanity code to distribute the published channel.
