It is used to play audio, video, or image files from a connected USB drive. (both thumb/flash drive and larger hard drives/SSD drives/NAS setups)

The content is then generally played through the Roku Media Player.

Is there a possibility that if I connect a pair of USB headphones to this port that I might be able to listen to the audio content streamed through the unit, just like it was being sent to the tv/monitor speakers thru the HDMI ?

No, it only support USB data.
Depending on your Ultra model number (and the remote you are using), you can either connect wired earphones to the remote (if it has a 3.5mm jack), or you can connect wireless headphones to either the Ultra directly (newer models), or use the official Roku mobile app that allows you to use your mobile device as a remote and provides Private Listening via bluetooth.

You might want to look on your TV as well for any headphone support jacks.
