RS232C port

Is there a way to set up my roku box using only a USB to rs232 db 9pin female serial cable?

No.  The USB port on most Rokus is for power only.  On the high-end models it's used for attaching a drive with your own media or a USB to Ethernet adapter on the Streambar/Soundbar.
