Suddenly my play/pause button no longer works. When I press it the instructions for using the voice button pops up instead of pausing or resuming play. All other remote functions are working fine. Tried new batteries, no difference.
Try:
1) Check for updates: Settings/System/System updates/Check now
2) Rerun remote setup: Settings/Remotes & devices/Remotes/<remote name>/Set up remote for TV control
3) Restart your Roku: Settings/System/System restart/Restart
Didn't help.
What happens when you press the mic/voice button?
It works normally. And right now is the only way I can pause or re-start playing after pausing.

Wait. What? Did you just say that the mic/voice button does pause/play? And you said mic/voice button also works normally? Those two things are not consistent.
Describe what happens when you press mic/voice, then describe what happens when you press pause/play.

I interpreted that mean to they have to use the "play" or "pause" voice command to play/pause (it works of course)
I'm trying to figure out the genesis of, explanation for, and resolution to this issue - its a weird one.

Try this:
1) Go to Settings/Privacy/Microphone/Channel Microphone access and switch it to "Never"
2) Go to Settings/Privacy/Microphone/Channel permissions/Reset channel permissions
3) Check for updates: Settings/System/System updates/Check now
4) Restart the Roku: Settings/System/System restart/Restart
Here is what appears when I press the pause/play button. The mic button (and all other buttons) acts normally. It allows me to speak commands.
That's very odd.
I have to ask one question, just to make sure. Are you using the remote that came with the device (or another Roku brand remote)? Or are you using a third party remote, such as Logitech or some other brand?
