Different channels on two devices

Is it possible to have channels on my Roku TV, upstairs, but not have them on the living room TV, in the living room?

Not if they're on the same Roku account. All devices tied to a single Roku account will have the same apps installed. Adding to one adds to all; removing from one removes from all.

If it's really important that the apps on the menu be different, you can set up a separate Roku account for the second device. There are some drawbacks to this. If you use Roku billing for subscriptions, that would cause an issue. If you subscribe directly to a service -- say, you pay Netflix directly for their service and didn't subscribe through Roku -- then there would not be that issue.
