RMP has not been updated in a number of years. My guess is it's indexing every file on the USB drive, which can take some time. You might instead consider using a media server that supports the DLNA protocol, which RMP supports. Serviio is one example, but there are many others, and most are available free of charge. It requires a computer running on your network, but the plus side is it's available to any device on your network that supports DLNA (most smart TVs do, as well as Blu Ray players and other streaming devices).

Let's see if this works for you:
- Unplug and replug the USB drive from your Roku Ultra
- Remove the Roku Media Player app
- Restart your Roku device
- Add again the Roku Media Player app
